Background and aim: Teratospermia, also known as teratozoospermia, is characterized by abnormal sperm morphology, where a considerable number of sperm exhibit structural abnormalities. Teratospermia can be considered as an indicator of male infertility, and genetic variants in key genes can increase its development. The purpose of this study was to investigate the correlation between the rs2231137 polymorphism of the ABCG2 gene and teratospermia. Materials and methods: A case-control study design was employed, comprising 73 individuals with teratospermia and a control group of 154 people and then semen samples were obtained from all participants. After DNA extraction, genotyping of the rs2231137 polymorphism was performed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: The rs2231137 genetic variation was examined in both the control and teratospermia groups. In the control group, the genotypic distribution consisted of 87.66% GG, 11.04% GA, and 1.30% AA, while the teratospermia group exhibited 78.08% GG, 15.07% GA, and 6.85% AA. As a final point, after statistical analysis, this genetic variation was found to be associated with an increased risk of teratospermia (OR= 5.92, 95%CI= 1.12-31.42, P= 0.037). Conclusion: Based on our findings, the rs2231137 polymorphism could be considered as a risk factor for teratospermia.
