Abstract

Introduction & Objectives: The recent decade it has been indicated that Reactive Oxygen Species (ROSs) play a role in various types of male infertility and it leading to decrease Total Antioxidant Capacity (TAC) in the reproductive system. Xenobiotics and ROSs has been shown to make biological effects via the aryl hydrocarbon receptor (AhR) signaling pathway. The AhR function is regulated by AhR repressor (AhRR) then, its variants may affect the pathway. The AhRR Gene c.565C>G Polymorphism results in Ala to Pro replacement in position 185 of AhRR protein that its may be associated with smokers and hookah pipe infertile men .TAC is biochemical marker of ROSs stress in cell. The aim of this study was to determine whether or not AhRR Gene c.565C>G Polymorphism has an impact on this marker in seminal of smokers and hookah pipe infertile men (as a patient groups) in comparison of control groups (non smokers and hookah pipe infertile men.) Materials & Methods: Forty seminal samples including: 22 normal samples, as a control group and 18 case samples, were collected from the Fatemezahra IVF centre (Babol, Iran). Seminal plasma TAC levels in all samples were measured by TBARs and FRAP methods, respectively. The c.565C>G Polymorphism were genotyped using PCR-RFLP method . Results: Our results showed that, AhRR Pro185Ala genotype was implicated in susceptibility to TAC biochemical marker of ROSs stress in seminal plasma. TAC level in seminal plasma of control groups was significantly higher than patient groups. Conclusion: Thus, it seems that the AhRR Pro185Ala polymorphism probability affect elevate total antioxidant capacity in seminal plasma of smokers and hookah pipe infertile men .
